Aging is associated with profound changes in immune cell function. To investigate how aging impacts plasmacytoid dendritic cells (pDCs), we profiled purified pDCs from healthy adult (20–40 years) and elderly (>65 years) human donors. Using bulk RNA-seq (GSE269411) and bulk ATAC-seq (GSE279911), we identified transcriptional and epigenetic changes that occur with age. These datasets provide a resource to explore age-associated alterations in pDC identity, gene regulation, and chromatin accessibility. pDCs were FACS-sorted from 6 adult (20-40 years old) and 6 elderly (>65 years old) donors directly into Qiazol lysis buffer (Quiagen) and frozen at -80C for RNA sequencing. RNA extraction and library preparation were performed by the Stanford Functional Genomics Core. Amplification and qPCR were performed simultaneously after all samples were collected. Quality control was conducted using the Bioanalyzer analysis through the Stanford Functional Genomics Core. Samples were barcoded and pooled for sequencing. Samples were sequenced on a NovaSeq 6000 through the Stanford Functional Genomics Core.
