Human SCLC (small cell lung cancer), H82 and H146 cells were cultured. H82 cells were used for checking DNMT1 localization at the proximal promoter region of NEUROD1 target genes by CUT&Tag (Cleavage Under Targets and Tagmentation) assay. H146 cells were treated with DNMT1 inhibitor and analyzed for ASCL1 target gene expression by RNA-seq analysis. DNMT1 repressed the neuroendocrine lineage-differentiation gene expression which can be reversed by using decitabine, the DNMT1 inhibitor.
