PAX8 knockdown was performed in uterine serous papillary carcinoma cells (USPC) to identify novel genes and cellular processes regulated by PAX8 in USPC. PAX8 was knocked down using lentiviral particles carrying one of two different PAX8 shRNAs, or a control shRNA in USPC-ARK-1 and USPC-ARK-2 cells, in triplicates. Cells carrying puromycin-resistant vector were then selected for 48h using puromycin, and the RNA was harvested 24h after selection and sent for next-generation sequencing.
