Hevea brasiliensis is an economically important tropical tree that produces natural rubber. Among diverse stress factors, low temperature is a key factor limiting the rubber plantation extending to high latitude area. Lower temperature is responsible for stopping latex production for 1 months per year in sub optimal areas. High attention have been paid to breeding cold tolerant cultivar of H. brasiliensis and many studies have focused on the altered physiological parameters under chilling stress condition, but the molecular mechanism of cold responses in Hevea brasiliensis has not been well investigated. In previous work, we have proved that epigenetic modification plays important role in cold acclimation of Hevea brasiliensis. In this work, reduced representation bisulfite sequencing RRBS analysis was used to study global DNA methylation status alteration in genome wide under cold stress in Hevea brasiliensis. Study result showed that cold treatment induced global DNA demethylation and altered the sequence contexts of mCpG islands. Digital Gene Expression DGE analysis showed that cold induced 3693 up regulated genes and 2043 down regulated genes. Integrative analysis of DNA methylome and trasncriptome revealed that transcriptomic changes of over 400 genes were associated with alteration of DNA methylation. Cold treatment reprogramed the methylation site of CpG islands in the genome of Hevea brasiliensis. Cold induced demethylation in upstream region showed close correlation with high gene expression. Indicating coordinated reprogramming of epigenome and transcriptome in Hevea brasiliensis after cold treatment.
