Dendritic cells (DCs) simultaneously prime and dampen the immune response. The transcriptional program that regulates immunosuppression in DCs is currently unknown. Here we identified ETS homologous factor as a key transcription factor that regulates DC immunosuppression. PBMCs of four male healthy volunteers were plated individually in complete RPMI-1640 (RPMI-1640 with 10% FCS, 1% L-glutamate, 1% penicillin/streptomycin, and 55 μM 2-mercaptoethanol), and stimulated with 100 nM TLRstimulation for 16 hrs. Then, DCs (Lin- CD123- CD11c+ HLA-DR+) were FACS-sorted and pooled for scRNA-seq library preparation.
