The study aims to investigate the impact of disruptive motion, specifically orbital shaking and magnetic stirring, on metabolic activities, degradation rates, and community composition of syntrophic acid-oxidizing cultures grown in high ammonia conditions (0.3M).The culture was grown batch-style subjected to either no-motion, orbital shaking and magnetic stirring and fed either 50mM acetate or 50mM propionate. Each experimental condition was run in triplicates, resulting in the following samples:CA1-4 - No motion, Acetate, replicate 1-3CP1-4 - No motion, Propionate, replicate 1-3StA1-4 - Magnetic stirring, Acetate, replicate 1-3StP1-4 - Magnetic stirring, Propionate, replicate 1-3ShA1-4 - Orbital shaking, Acetate, replicate 1-3ShP1-4 - Orbital shaking, Propionate, replicate 1-3Two types of sequencing was performed during the study and uploaded to this project:16s gene amplicon sequencing:Sampled periodically throughout the degradation for both acetate- and propionate-fed cultures.metatranscriptomic sequencing:Sampled for both acetate- and propionate-fed cultures during the exponential phase of acid degradation, after substrate had been added a second time.The extracted RNA was merged for the acetate-fed replicates, resulting in a total of 3 samples(CA,StA,ShA), one for each motion. Propionate-fed replicates were not merged, resulting in a total of 9 samples (CP1-3,StP1-3,ShP1-3)
