Data for Mendonca et al., 'The Mitotic Chromosome Periphery Modulates Chromosome Mechanics' (2025) Two datasets are included: Raw data from stretch-hold experiments for microrheology analysis Experiments were performed at 100 um/s ('WT_100umps', 'KD_100umps' and 'OE_100umps') or 0.2 um/s ('WT_0.2umps') on individual wild-type ('WT'), Ki-67 knock-down ('KD') or over-expression ('OE') chromosomes. Data were generated using a LUMICKS c-trap optical tweezer instrument. *.h5 file structures contain experimental parameters: Force, Distance and Time, at high frequency along with experiment metadata and were exported from Bluelake version 2.5.1 (LUMICKS). A detailed explanation of the experimental procedure can be found in the paper.These data can be processed to extract complex moduli for individual chromosomes using custom scripts that can be accessed at: https://doi.org/10.5281/zenodo.14527725 Burgers model fits to complex moduli Microrheology data from individual chromosomes were fit to an equation for the Kelvin representation of the Burgers Model (see Mendonca et al., 2025 for details) by minimising the error function while solving a nonlinear least squares problem in MATLAB to extract elastic and viscous components of chromosome mechanical properties.
