AST- and NAS-derived dopamine neurons were compared for their gene expression profiles after differentiation. Functional data was also collected in terms of neurophysiology. Overall design: AST- and NAS-derived dopamine neurons were compared for their gene expression profiles after differentiation. Functional data was also collected in terms of neurophysiology. iPSCs were generated from a member of the Iowa kindred carrying a triplication of the SNCA locus (AST) and an unaffected member of the family without this mutation (NAS). Dopamine neurons were differentiated from iPSCs. Transcriptome profiling was done on n=3 AST and n=3 NAS (replicate and independent cell cultures).
