Cytoplasmic DNA Sequencing of THP-1 cell lines

THP-1 cells were observed to induce interferon-stimulated gene (ISG) expression upon knock-out of SAMHD1 in a cGAS-dependent manner. Given that cGAS detects and responds to cytoplasmic DNA, we performed cytoplasmic DNA sequencing in THP-1 cells with and without SAMHD1. Parental THP-1 cells and three monoclonal SAMHD1 knock-out THP-1 cell lines were subject to cytoplasmic-nuclear fractionation. DNA was extracted from the cytoplasmic fraction of each cell line and used directly for deep sequencing on the Illumina platform.
