We propose a strategy to boost the therapeutic efficacy of Oncolytic therapy by combining it with immuno-modulating agents targeting protein tyrosine phosphatases. The mechanism of action was examined by microarray analysis using the Affymetrix Human PrimeView Array. 786-0 cells were plated in 6-well dishes and allowed to adhere overnight. The next day, cells were pretreated for 4hours with orthovanadate (150uM), metavanadadate (150uM) or the vehicle. Following pre-treatment, the cells were infected with VSVΔ51 at an MOI of 0.01 or left uninfected. Twenty-four hours post infection, RNA was collected using an RNA-easy kit . Biological triplicates were subsequently pooled and RNA quality was measured using Agilent 2100 Bioanalyzer (Agilent Technologies) before hybridization. Hybridized to Affymetrix Human PrimeView Array . Microarray data was processed using Transcriptome Analysis Console (TAC) 3.0 under default parameters of Gene Level Differential Expression Analysis. Fold change in gene expression was calculated for each gene in relation to uninfected, untreated control.
