Whole-genome sequencing of phazolicin-resistant Sinorhizobium meliloti mutants

To identify the transmembrane transporters involved in the internalization of azole-modified antibiotic phazolicin the Tn-library was prepared using the phazolicin-sensitive model strain Sinorhizobium meliloti 1021. Four selected mutants, which demonstrated the growth on the medium containing 20 mkM phazolicin were subjected to whole-genome shotgun sequencing to identify the genes disrupted by the transposon insertion and other mutations accumulated in the genome. All four mutants had mutations in the genes bacA and yejABEF encoding to unrelated peptide transporters.
