By using oligonucleotide microarray-based technology as a tool for rapid mapping of unique differences in gene expression, we found that inhibiting serpin B13 in BALB/c mice treated with a mAb according to our protocol resulted in the upregulation of relatively few genes, including several Reg genes, which have been implicated in β-cell proliferation. Overall design: BALB/c mice were intraperitoneally injected four times with 25 ug of serpin B13 mAb (clone: B29) or control mAb (IgG) (total dose of 100 ug) and sacrificed two weeks after the last injection.The islets were isolated and separated according to their diameter (200 μM). Total RNA was isolated from the islets. The results between serpin B13 mAb and control mAb were compared in the islets of different sizes.
