By combining CTAB extraction buffer and lab-made SA-coated magnetic nanoparticles, we tested a quick and inexpensive high-throughput method to isolate quality genomic DNA from two different tissues of three crops; maize (Zea mays), soybean (Glycine max) and wheat (Triticum aestivum) in less than 2 hours with fewer steps than a standard CTAB extraction method. The isolated DNA was proved to be of sufficient quality and quantity in downstream application in multiple molecular breeding tools, such as polymerase chain reaction (PCR) and molecular inversion probe (MIP).
