We performed gain-of-function (GoF) and loss-of-function (LoF) analyses of Midkine in melanoma cells by RNA-seq. Overall design: For LoF, midkine was depleted in SK-Mel-147 (a melanoma cell line with high midkine levels and inherent metastatic potential) using MDK-sh5 (NM_002391.3-621s21c1). In turn, GoF studies were performed by overexpressing midkine in non-metastatic WM164 melanoma cells using ORF lentiviral expression vector pReceiver-Lv105-A0792 (MDK) .
