Innate lymphoid cells (ILCs) and interleukin-22 (IL-22) are protective against C. difficile infection. We sought to therapeutically stimulate IL-22 production from ILCs to protect against infection. Oral administration of TLR-7 agonist R848 protects mice against lethal C. difficile infection without altering intestinal microbiome. R848 stimulated IL-22 secretion from ILCs and limited toxin-mediated intestinal damage.
