Methane is an important greenhouse gas, methanotrophs play an important role in CH4 cycle as the CH4 sink. With the global warming, the hydrological process of glacier melting was influenced and the meadow in glacier foreland was transformed into swamp meadow. while the response of methanotrophs to the glacier melting was not clear. Tibetan plateau has the most glacier except polar regions, hummock and hollow are common microtopography in Tibetan plateau. laboratory culture of soil from different microtopography in Tibetan glacier foreland experiment was conducted to study the methanotrophic community under different temperature and moisture in total DNA (pmoA and 16S rRNA) and 13C-labeled DNA (16S rRNA) by using qPCR, DNA-SIP and high-throughput sequencing.
