This project aims to investigate the transcriptome changes in the hippocampus and prefrontal cortex following cecal ligation and puncture (CLP) surgery (a well-established animal model for simulating sepsis) with Shenfu Huang (a traditional Chinese medicine compound) treatment. The study focuses on identifying differentially expressed genes and exploring potential molecular mechanisms underlying sepsis-induced neurological alterations, as well as the regulatory effects of Shenfu Huang, in these two critical brain regions.Sample collection and processing: Total RNA was extracted from the hippocampus and prefrontal cortex tissues of animals after CLP surgery with Shenfu Huang treatment. RNA integrity and purity were rigorously assessed using two complementary instruments: NanoDrop 2000 spectrophotometer (Thermo Fisher Scientific, USA) for measuring OD260/280 ratio (1.8-2.2, indicating high purity) and Agilent 2100 Bioanalyzer (Agilent Technologies, USA) for determining RNA Integrity Number (RIN >= 6.5, ensuring good integrity). Only high-quality RNA samples meeting these criteria were selected for subsequent library construction.
