We developed a 454 cDNA amplicon sequencing method for alpha-gliadin transcripts encompassing the three major CD epitopes present in alpha gliadins, and used it to test a panel of wheat plants (n=77) derived from 61 different durum wheat cultivars and accessions. A dedicated sequence analysis pipeline returned a total number of 304 different alpha-gliadin nucleotide sequences resulting in a library of 171 deduced “unique deduced protein fragments” of alpha-gliadins which was used for profiling the CD epitopes expressed in the endosperm of wheat plants quantitatively and qualitatively. A few plants revealed lower expression of CD epitopes, but none free of CD epitopes.
