The invention describes the variation of the apparent Michaelis–Menten constant (Km) as a function of enzyme concentration. The phenomenon may occur in both Michaelis–Menten–type enzymes and allosteric enzymes; however, in allosteric enzymes, increasing the enzyme concentration raises the apparent Km value by decreasing the apparent substrate–enzyme affinity, which may lead to the blockage of the metabolic flux in pathways where the enzyme represents a regulatory point. This property enables modulation of allosteric enzyme activity by adjusting enzyme concentration, offering a method for intervention in metabolic pathway regulation, inhibitor design, and applications in pharmacology and enzymatic therapy.
