A Sleeping Beauty screen to identify gain- and loss-of-function modifications that enhance T-cell infiltration into tumors

One challenge limiting T cell-mediated cancer immunotherapy success is that tumors lacking T-cell infiltration respond poorly to treatment. We performed a forward genetic screen in mice that used the Sleeping Beauty (SB) mutagenesis system to identify candidate genes in T cells that might be modified to drive intratumoral T-cell accumulation. The data deposited include SB insertion site sequence data from all tumor models, prepared using an LMPCR approach to amplify the transposon and adjacent genomic sequence.
