Transcriptome analysis of 7-day-old Marchantia WT gemmalings and MprbohA rbohB mutant cell masses

NADPH oxidases (NOXs), also known as Respiratory Burst Oxidase Homologs (RBOHs) in plants, produce reactive oxygen species (ROS) in many eukaryotes. We generated loss-of-function mutants for both RBOH genes (MpRBOHA and MpRBOHB) in the liverwort Marchantia polymorpha. The MprbohA rbohB double mutant exhibits severe developmental defects, resulting in undifferentiated cell masses. To investigate this phenotype, we compared gene expression profiles between 7-day-old WT gemmalings and mutant cell masses. Plants were cultured in half-strength Gamborg's B5 liquid medium containing 1% sucrose under continuous white light for 7 days prior to RNA extraction.
