This project investigates the impact enodcrine disrupting chemicals (EDCs) on differentiation toward stem cell derived islets. RNA sequencing (RNA-seq) was performed on EDCs treated and non-treated islets differentiated from iPSCs to SC-islets. By comparing the transcriptomic profiles of these islets, the study aims to uncover molecular mechanisms affected by EDCs treatment, and the impact on SC-islet fate. Overall design: iPSCs were differenitated toward ST-islet fate using a 7 stage step wise protocol. At Endocrine progenitor stage the each well of aggreagtes were split in 2 wells, one for treatment and one for control. For 14 days the treatment were treated with a 1 mM mix of bisphenol A, bisphenol S and trans-nonachlor. The SC-islets were harvested at day 38 of the protocol, 7 days after end of treatment.
