Germline mutations in the RNA Helicase gene DDX41 cause inherited predisposition to Myelodysplastic Syndrome and other myeloid blood malignancies. We characterized the effect of loss of one or both copies of the Ddx41 gene on the gene expression profile of immortalized hematopoietic progenitor cells. Overall design: To determine the role effect of loss of Ddx41 on hematopoietic progenitor cells, we generated immortalized hematopoietic cell lines from Ddx41-floxed; Rosa-CreERT mice. This approach allows for the inducible deletion of Ddx41 and timecourse analysis of the cellular response to loss of Ddx41. Since loss of both alleles of Ddx41 causes cell death by 72hours post-excision, we characterized the gene expression of the cells at the 48 hour timepoint after induction of Cre activity by treatment with tamoxifen.
