Conclusion:
Regarding the CD8-negative or double-negative (CD4- CD8-) population observed in the 2C panel, this subset likely represents 2C TCR-expressing cells that have downregulated or lost CD8 expression. Notably, this population was present from day 0, prior to adoptive transfer, despite employing the same CD8+ naïve purification kit that was utilized for OT-I cells. This observation suggests an inherent characteristic of the 2C TCR cell population rather than an artifact of the experimental procedure.

2C TCR cells are identified using the clonotypic antibody 1B2, which is specific for the 2C T cell receptor. The 1B2 antibody (clonotypic) recognizes an epitope that spans all six complementarity determining regions (CDRs) of the 2C TCR. This unique epitope makes 1B2 highly specific for the 2C TCR, allowing for precise identification of these cells. Cytotoxicity
