• IGF2BP1 promotes leukemia stem cell phenotype by regulating HOXB4, MYB and ALDH1A1.• Inhibition of IGF2BP1 reduces leukemia tumorigenicity, and enhances leukemia cell death and differentiation. RNA sequencing analysis of 697 (EU3) ALDH+ subpopulations with and without IGF2BP1 knockdown was performed from four biological replicates (n=4). Each biological replicate represents de novo selection of ALDH+ cells and independent doxycycline treatments.
