We explored the mechanism of WNT16 on LESCs by performing RNA-sequencing and analyzing differentially expressed genes (DEGs) between WNT16-treated LESCs and controls. CXCR4 was identified as a vital factor in response to WNT16. To explore the downstream signals of WNT16/CXCR4, key proteins involved in several potential signaling pathways were detected. The activation of CXCR4/MEK/ERK signaling was found to play a pivotal role in mediating the effect of WNT16 on LESCs.
