IgNAR exhibits significant promise in the fields of cancer and anti-virus biotherapies. Notably, the variable regions of IgNAR (VNAR) possess comparable antigen binding affinity with much smaller molecular weight (~12 kDa) compared to IgNAR.Next-generation sequencing (NGS) will be another method for VNAR screening with a number of advantages.Sequencing the obtained VNAR libraries through NGS can quickly and efficiently obtain immune-specific sequence information, which provides a basis for further screening of specific high-affinity antibodies. Following eight doses of immunization with Cre, Cre_NT, and BSA antigens, total RNA was isolated from the spleens and PBMCs of the sharks, and reverse transcribed into cDNA. The VNAR library was then amplified using white-spotted bamboo shark-specific VNAR primers.
