Transcriptomic Analysis of Myelodysplastic Syndrome Mouse Models Treated with Polyphenolic Compounds by RNA-Seq

Myelodysplastic syndrome (MDS) is a heterogeneous group of hematological disorders characterized by ineffective hematopoiesis, cytopenias, and a high risk of progression to acute myeloid leukemia (AML). Experimental models are essential to understand the molecular mechanisms underlying the disease and to evaluate potential therapeutic strategies. In this study, we used transgenic female mouse models of MDS (C57BL/6-Tg/Vav1-NUP98/HOXD13 and BALB/c-PML/RARA) to investigate the transcriptomic effects of polyphenolic compounds with anti-inflammatory and antioxidant properties. RNA-Seq analyses were performed on bone marrow and spleen samples from female NUP98 mice treated with epigallocatechin-3-gallate (EGCG), as well as spleen samples from female BALB-PML and NUP98 mice treated with curcumin or quercetin. Bioinformatic analysis identified distinct and tissue-specific gene expression profiles associated with each treatment, revealing modulation of metabolic, immune, and signaling pathways relevant to disease progression. This study provides insights into the molecular impact of natural polyphenols in MDS models, contributing to the understanding of their therapeutic potential and their role in the regulation of hematopoietic and leukemic processes.
