This study was designed to evaluate the lung transcriptome analysis against dexamethasone treatment throughout HDM challenge period in the aged (80-82 Weeks) mice. The lung samples from aged inflammed mice with or without dexamethasone treatmrnt were isolated followed by RNA isolation and RNA sequence analysis. Overall design: First, we sensitized aged (80-82 Weeks) mice with 50 μg of HDM in a volume of 40 μL through intranasal (i. n) route on days 1, 3, and 5. The challenge phase consisted of 3 consecutive phases of 3 doses (i.n) of HDM (50 μg) on days 11-13, 18-20, and 25-27 respectively. Dexamethasone (4 mg/Kg body weight, intraperitoneal, route) treatment was given throughout the challenge period after every 72 h interval. Post 24 h, last challenge all the lung samples were collected for futher processing. Further, RNA was isolated, checked for RNA quality and processed for RNA sequence analysis.
