To investigate the molecular mechanism by which HEY1/ETS1-B16F10 niche cells enhance CTL function, we performed RNA-seq analysis using mCherry expressing cells as negative controls. B16F10-0VA, mCherry,Hey1,Ets1, Hey1+Ets1 weretransfected and 1x10 cells were plated in a 6-well plate, cells were treated with 1 ng/mldoxycycline in complete medium DMEM every 24h and collected 96h after plating. All cellsamples were lysed with TRlzol reagent, rapidly frozen on dryice,and shipped to the sequencing company for analysis.
