Shown are SCAR null cells expressing GFP-WASP (bottom half of the image) and ArpC4-mRFP (top half of the image). Cells were grown overnight in low fluorescent medium (LoFlo) to reduce autofluorescence. Cells were harvested from the dish and resuspended in LoFlo. 100 ul of cell suspension was pipetted onto a glass bottom dish that contained 2 ml of LoFlo medium.
TIRF microscopy. Nikon 1.45 NA 100x Plan Apo TIRF objective. 50 ms illumination per frame. 1 frame per 2 seconds. A beam splitter was used to project the mRFP signal to the top half of the camera and the GFP signal to the lower half of the camera.
