This study is investigating the genetic events required for renal pathogenesis in the mouse kidney using an unbiased Sleeping Beauty transposon mutagenesis model. Mice heterozygous for a renal specific CreERT2 allele (PaxACreERT2 transgene), floxed Vhl allele, Rosa26-SB13 (transposase) and T2Onc (transposon) alleles are induced with tamoxifen and observed for tumour induction. Tumour DNA is being sequenced for identification of common mutations (transposon insertion sites or other genetic events). Correlation of identified mutations with known mutational events in human kidney cancer will help identify the events required for carcinogenesis within the mouse kidney.
