To understand the effect of age on miRNA profiles in male mouse vertebrae, we isolated L4-5 vertebral samples and digested with endotoxin-free collagenase (Liberase) twice for 30 minutes. The resulting bone was confirmed to be largely osteocyte-enriched and RNA was isolated using QIAzol reagent. Overall design: We then performed miRNA gene profiling between the young (6 month) and old (24 month) vertebral samples.
