To explore the role of auxin in Ceratopteris richardii root initiation, we carried out RNA-seq to analyze gene expression levels upon 2,4-D treatment. Ceratopteris richardii (wild type RNWT1) spores were cultured on 1/2MS medium at 26 °C under a 16-h-light/8-h-dark photoperiod in a growth chamber. After 30 days (mock group), sporophyte seedlings were transferred to medium with 2 μM 2,4-D for 24 or 48 h. The whole seedlings were harvested and frozen in liquid nitrogen for RNA extration. Library construction and deep sequencing were carried out using the Illumina HiSeq 3000 platform following the manufacturer's instructions by Genergy Biotechnology (Shanghai, China).
