We used mice as an experimental model to establish two animal infection models of bacteremia with different outcomes. In the low-dose infection group, the bacteria within the mice are cleared by the immune system, allowing the mice to survive. In contrast, the high-dose infection group leads to sepsis and ultimately results in the death of the mice. We aim to utilize single-cell sequencing technology to study the changes in the transcriptomes of various immune cells during different stages and outcomes of mammalian bacteremia, in order to understand the host's response to bacteremia. Overall design: To collect PBMC from animals in lethal and non-lethal bacteremia models for scRNA-seq, groups of 6 mice were infected with E.coli at high (lethal dose, H group) and low (non-lethal dose, L group) doses, respectively. For L group, four time points, 2, 4, 8, and 12 hpi, were selected to indicate different stages of infection; while for the high dose infection group, two time points, 2, and 6 hpi, were selected to indicate early and late moribund stages of infection.Isolated PBMC were then subjected to scRNA-seq with 10Ã Genomics platform.
