Here we report the effects of Active DNA-dependent ATPase A Domain inhibitor (ADAADi) on human transcriptome Overall design: HeLa cells were maintained in Dulbecco's Modified Eagle Medium (DMEM) supplemented with 10% fetal bovine serum (FBS), 1% penicillin-streptomycin-amphotericin cocktail at 37°C in the presence of 5% CO 2 . Cells were treated with 5mM of ADAADi and incubated for 48 hours at which cells were harvested and total RNA was purified using Trizol (Sigma Aldrich, USA). Poly(A) mRNA was purified from total RNA using NEBNext® poly(A) mRNA magnetic isolation module (Catalog # E7490S), with certain optimization in the user protocol provided by the manufacturer. RNA-seq library was prepared using 100 ng purified poly(A) mRNA using NEBNext Ultra II RNA Library Prep Kit with certain optimization in the user protocol provided by the manufacturer.
