This study used CUT&Tag to profile the genomic binding sites of PARIS in hippocampal tissue from CamK-PARIS transgenic mice. The tissue was fixed, processed for CUT&Tag, and sequenced as paired-end libraries. Signal tracks and SEACR peak calls were generated to characterize PARIS-associated genomic regions and to relate PARIS binding to STAT3 signaling and downstream transcriptional changes in the context of neurodegeneration. Overall design: CUT&Tag libraries were prepared from hippocampal tissue of CamK-PARIS mice using an anti-FLAG antibody, along with an IgG control. Paired-end sequencing was performed on an Illumina platform. Reads were aligned to the mouse reference genome using Bowtie2, followed by filtering, SEACR peak calling with the IgG control track, and generation of bigWig signal tracks. Processed data include normalized coverage tracks and peak BED files.
