GFP and Flpo MEFs are mixed at 1:1 ratio and cultured on the same dish. Cells are treated with 10nM Thapsigargin for 16hr. Cells were then trypsinized and sent for scRNA sequencing. Overall design: CHOP FLuL/FLuL primary MEFs are infected with Ad-GFP or Ad-Flpo-GFP and GFP positive cells were sorted out by FACS and expanded. GFP cells do not express CHOP but express nanoluciferase instea whereas FLpo cells express CHOP but not nanoluciferase. GFP and FLpo cells were mixed at 1:1 ratio and plated in the same culture dish. Cells were then treated with 10 nM ER stressor thapsigargin for 16 hrs and collected for scRNA-seq.
