We used low-throughput (screening) data to investigate how ancient DNA (aDNA) from herbarium specimens degrades over time. We analysed 573 historical herbarium samples collected from seven herbaria across Europe and North America, focusing on the genera Hordeum (barley) and Oryza (rice). Four key metrics were used to assess DNA damage: endogenous DNA content, fragment length, cytosine deamination, and damage fraction per site.
