In this experiment, a heterotrophic nitrification-aerobic denitrification (HN-AD) strain with low-temperature tolerance and high-salt tolerance was screened from the activated sludge in the aerobic section of a domestic sewage treatment plant in Inner Mongolia. Through morphological characteristics and 16S rDNA analysis, it was identified and named as Pseudomonas knackmussii L4. The strain L4 can effectively metabolize ammonia nitrogen, nitrate nitrogen and nitrite nitrogen under the conditions of a temperature of 10 deg C and a salinity (calculated as NaCl) of 50 g/L. Genomic sequencing of the strain detected enzymes related to nitrogen metabolism such as periplasmic nitrate reductase and nitrite reductase associated with HN-AD, as well as their coding genes. Subsequently, the metabolic pathway of HN-AD was speculated. The results showed that Pseudomonas knackmussii L4 exhibited excellent nitrogen removal performance in the treatment of actual wastewater, which has broad application prospects in the fields of treating low-temperature and high-salt wastewater.
