We have described a facile high throughput yeast surface display coupled to FACS method to quantify binding affinity in large number of variants of an intrinsically disordered bacterial protein (IDP), CcdA. We also describe development of a predictive model for estimation of binding affinity changes upon mutations in IDPs.
