CHMP7/ESCRT-III is localized at the nuclear envelope of cortical neurons and required for expression of activity-regulated genes

The goal of this project is to analyze the effect of shRNA-mediated knockdown of Chmp7 on the transcriptome of primary cortical neurons. Murine primary cortical cultures were transduced on DIV 6 with AAVs encoding either Chmp7-targeting shRNA or a scrambled shRNA control. On DIV 14 cortical neurons were first pre-treated with NBQX for 16 h and then treated with bicuculline for additional 1 h. Following bicuculine stimulation, bulk RNA sequencing analysis was performed to compare transcriptomes of control vs Chmp7 knockdown cultures.
