This experiment is assocaited with a publication defining a tumor supressor role of FBXO11 in the transformation of MDS. We observe decreased expression of FBXO11 is associated with transformation to secondary AML. Overall design: This experiment was designed to determine the genes regulated by FBXO11. We compared 4 cell populations. 1. MDS-Cas9 (parent MDS-L cells expressing Cas9 - effectively a control), 2. MDSL-Cas9-FBXO11KO (MDS-L cells with Cas9 and FBXO11 sgRNA - effectively FBXO11 knockout), 3. MDSL-Cas9-FBXO11KOv1OE (FBXO11 knockout cells with overexpression of FBXO11 isoform 1 cDNA), and 4. MDSL-Cas9-FBXO11KOv4OE (FBXO11 knockout cells with overexpression of FBXO11 isoform 4 cDNA)
