E. coli is a well studied organism. Here we build a system to study the structure of a single activated transcription unit within the E. coli chromosome. We used HiC, RNA-seq, T7 RNA polymerase ChIP-seq and GapR ChIP-seq data to characterize the activation of the single transcription unit. The HiC data allow us to have the spatial organization of the single transcriptional unit, which correlates well with the RNA-seq and ChIP-seq of the T7 polymerase. Finally the GapR ChIP-seq allows us to have the negative supercoiling around the transcription uint.
