Effects of TCF-1 de-repression on heterogeneity of effector and memory progenitor T cell transcriptomes in acute infection [scRNA-seq]

The transcription factor TCF-1 is downregulated during effector differentiation of CD8+ T cells. However, how its downregulation impacts gene expression in different effector- and memory-biased populations remains unclear. Here, we generate mice in which TCF-1 fails to become downregulated in differentiated effector cells and probe the transcriptional impact of TCF-1 retention in different T cell differentiation states by scRNA-sequencing. CD8+ T cells specific to the LCMV-gp3 antigen were sorted from the spleens of two WT control and two Tcf7∆+22kb mice on 8 dpi with LCMV-Armstrong. Samples were subjected to scRNA-sequencing.
