In this study we analysed the functional relevance of the interaction between Pla1 and the MTREC complex core component Red1 using various high throughput sequencing analyses performed on different Pla1-Red1 interaction mutants.Our analyses revealed that as part of MTREC complex, Pla1 hyper-adenylates Cryptic Unstable Transcripts (CUTs) for their efficient degradation. Interestingly, our H3K9(me)2 ChIP seq. data analysis also showed us that a functional Red1-Pla1 interaction was also required for the efficient assembly of the fission yeast facultative heterochromatic islands. Together, our data suggest a complex interplay between the RNA surveillance and 3’-end processing machineries. Overall design: Detailed analysis of two Pla1-Red1 interaction mutants namely red1W298A/F305A and red1△288-345 with WT and total red1△ control strains using high throughput poly(A)+ RNA sequencing, total RNA sequencing, RNA-immunoprecipitation (RIP) followed direct RNA sequencing and ChIP seq. analysis.
