Haploid genome activation exposes genes with a specific chromatin signature for pollen selection

Here, we obtained datasets of single cell and/or cell type transcriptomes and epigenomes of the rice male germ line isolated at the different developmental stages from hybrid and/or inbred varieties. Analysis of the data pinpoints to a premeiotic set-up of a chromatin state to instruct male germ cell-type specific gene expression during the gametogenesis, reveals that the male haploid genome activation occurs shortly after meiosis, and uncovers a specific chromatin signature for haploid gene expression which has implication in haploid and pollen selection. Germ cells were obtained by micromanipulation. DNA methylation and transcriptome libraries were constructed by scBS-seq and scRNA-seq techniques. Histone modification library was constructed using CUT&Tag technology.
