Prostate cancer cell lines resistant to docetaxel induced by gradient concentration

We added docetaxel to the culture medium of prostate cancer cell lines PC3 and DU145, and gradually increased the concentration. After a long time of culture, we obtained a docetaxel resistant prostate cancer cell lines. Overall design: Gradient concentration induction was used to culture PC3 and DU145 resistance cell lines to docetaxel. The concentrations of docetaxel started at 10nm/ml for DU145 and 0.5nm/ml for PC3. Double the concentration of docetaxel in the medium when cell division was vigorous till the IC50 of DU145 come to 1800nm/ml and 15nm/ml for PC3. Named the resistance cell lines with DU145-R and PC3-R. All cells were cultured in a humidified incubator with 5% CO2, 100 units/mL penicillin, and 100 mg/mL streptomycin.
