Transcriptomic changes in ER-HOXB8 mouse cell line on Phf6 knockout

PHF6 is a transcriptional regulator mutated in 3-5% of acute myeloid leukemia. To understand the effect of loss and clinical mutation of PHF6 on the transcriptome of myeloid cells we generated multiple wildtype and Phf6 knockout clones. We observed that loss of PHF6 upregulated genes related to stemness and downregulates genes related to myeloid differentiation. RNA-Seq of wildtype and Phf6 knockout ER-HOXB8 mouse GMP cells that were clonally selected and expanded after Cas9-gRNA lentiviral transduction.
