All animal studies were conducted in accordance with the Guide for the Care and Use of Laboratory Animals of the NIH. Mice were maintained according to the guidelines of the Joslin Diabetes Center (JDC) Committee on Animal Care, and study designs were approved by the JDC Committee on Animal Care (protocol # 00-02). All mice were maintained in C57BL/6 background and housed in a temperature-controlled environment with 12 hr light/dark cycles. Muscle specific hsf1 knockout mice (HSF1MKO ) were generated by crossing HSA-Cre mice with the hsf1 flox/flox mice (HSF1F/F).For fed-fast-refed experiment, male mice at 8-10 weeks of age were fed ad libitum, or deprived of food overnight (15hrs), or refed with food for 3 hours after overnight food deprivation. Mice were sacrificed by CO2 exhalation and gastrocnemius muscle were collected immediately and snap frozen to keep. Mouse muscle RNA was isolated using the RNeasy Fibrous Tissue Mini Kit (Qiagen, 74704). Next-Generation RNA Sequencing was performed with illumina HiSeq 2500 and analyzed at Center for Cancer Computational Biology (Dana-Farber cancer institute, Boston).
